11月8日,由我校昆虫科学研究所刘树生教授作为第一作者兼通讯作者的学术论文《非对称交配互作驱动一种粉虱的广泛入侵及对土著生物的取代》在《SCIENCE》上发表,该杂志已于8日在北京举办新闻发布会。

B 型烟粉虱是全球100种最危险入侵生物之一,包括许多不同的遗传群或“生物型”,可能起源于地中海—小亚细亚地区,并且传播到世界大多数地方,被认为是世界上危害性最大的农业害虫。90年代初期入侵到澳大利亚,90年代中期入侵到中国。B型烟粉虱入侵后,种群迅速增长并扩张,有时候,一片棉花叶上会有上万只B型烟粉虱。这些害虫大量取食,危害番茄、烟草、棉花等数十种重要经济作物,传播双生病毒引发植物病毒病蔓延成灾,取代危害性不大的土著烟粉虱导致本地生物灭绝,所到之处农作物严重减产甚至绝收,被国际科技界称为“超级害虫”。目前正给中国的农业和环境造成很严重的破坏。

为了探索其“超级入侵”本能的奥秘,世界各国的科学家进行了长期的努力,但一直都没有找到答案。

刘树生教授领衔的科研团队于1995年-2006年间在中国浙江、澳大利亚昆士兰共40多个地点进行野外系统采样调查。通过长期研究课题组发现,B型烟粉虱之所以能让成片的农作物绝收,因为它有两大“武器”:一方面利用交配行为入侵,让土著烟粉虱绝种;另一方面与病毒强强联合,既加快自身的繁殖,又加速植物病毒病的流行。 “非对称交配互作”是B型烟粉虱入侵的一个关键机制,它在促进B型烟粉虱数量增长的同时压抑土著烟粉虱种群增长,促进B型烟粉虱迅速入侵和扩张,取代危害性不大的土著烟粉虱。B型烟粉虱与它所传播的植物双生病毒(对植物危害最严重的一类病毒)之间存在互利共生关系,这种关系既有利于B型烟粉虱的繁殖入侵,又加速了病毒病的流行,经B型烟粉虱传播后,植物双生病毒的“毒性”也越来越强。这项由浙江大学昆虫科学研究所、中国农业科学院植物保护研究所与澳大利亚同行合作的研究成果,揭示了B型烟粉虱产生强大入侵能力的行为机制。

《科学》杂志论文评审专家在阅读了刘树生等投寄的论文后,对其做了高度评价。他们认为,这一研究综合应用生态学、行为学、分子生物学等领域的方法,将广泛系统的野外调查、巧妙的实验种群试验和独特且详细的行为观察有机地结合应用,而且研究在位于中国和澳大利亚的两个地理区同时进行,设计之巧妙,数据之系统和完整,在入侵生物学研究领域实属罕见,所做的结论令人信服。他们指出,该研究是入侵生物学领域的开拓性工作,将对这一学科的发展起到显著的推动作用,同时昆虫学、农学、动物行为学、进化生物学等学科领域也会从中得到启迪。美国、英国、德国等世界各国的媒体记者近几天纷纷通过各种方式采访刘树生教授。(农学院)

Carbon dioxide (CO2) is an important environmental cue for many organisms but is odorless to humans. It remains unclear whether the mammalian olfactory system can detect CO2 at concentrations around the average atmospheric level (0.038%). We demonstrated the expression of carbonic anhydrase type II (CAII), an enzyme that catabolizes CO2, in a subset of mouse olfactory neurons that express guanylyl cyclase D (GC-D+ neurons) and project axons to necklace glomeruli in the olfactory bulb. Exposure to CO2 activated these GC-D+ neurons, and exposure of a mouse to CO2 activated bulbar neurons associated with necklace glomeruli. Behavioral tests revealed CO2 detection thresholds of approximately 0.066%, and this sensitive CO2 detection required CAII activity. We conclude that mice detect CO2 at near-atmospheric concentrations through the olfactory subsystem of GC-D+ neurons.

Dopamine-Mushroom Body Circuit Regulates Saliency-Based Decision-Making in Drosophila

Drosophila melanogaster can make appropriate choices among alternative flight options on the basis of the relative salience of competing visual cues. We show that this choice behavior consists of early and late phases; the former requires activation of the dopaminergic system and mushroom bodies, whereas the latter is independent of these activities. Immunohistological analysis showed that mushroom bodies are densely innervated by dopaminergic axons. Thus, the circuit from the dopamine system to mushroom bodies is crucial for choice behavior in Drosophila.

自中国科学院物理研究所的汪卫华研究组在非晶合金的塑性研究中取得的重要进展。他们成功研制出在室温条件下同时具有超高
塑性和超高强度的块体非晶合金材料,并对其进行了结构分析,提出了探索塑性大块非晶的新方法,在物理上证明高强度和大塑性(结构材料最重要的两个性质)在非晶材料中可以有机结合在一起。此外,该材料的组元均是价格较低廉的常见金属,结合其优良的性能,使得该非晶合金有望得到广泛的实际应用。

这一研究组几年来在非晶态合金材料的塑性变形方面做了较系统的研究,澄清了脆性材料断裂研究中一些重要科学问题,加深了对非晶态等脆性材料的断裂机制的认识【见Phys. Rev. Lett. 94, 125510(2005) 】,在此基础上提出了切变唯像模型,利用泊松比有效地标定了大部分大块非晶合金的室温塑性【见Philo. Mag. Lett. 85 (2), 77-87(2005),, J Appl. Phys. 99, 093506(2006)】为改进非晶材料的脆性,设计新的具有大塑性、高强度的金属玻璃材料提供了必要的理论依据。并在此基础上首先开发出了具有较大塑性的CuZr基单相非晶合金材料【见Phys. Rev. Lett. 94(2005)205501】。

北京生命科学研究所马力耕博士实验室在3月8日《SCIENCE》杂志网络版上发表论文,报道一种G蛋白偶联受体是植物激素脱落酸的受体,该受体通过与异三聚体G蛋白α亚基直接相互作用传递脱落酸信号。论文的第一作者是刘西岗,其他作者还有岳艳玲、聂艳丽、李薇和中国农业大学的李斌和武维华博士,马力耕是通讯作者。

2007年3月8日,北京生命科学研究所马力耕博士实验室在科学(SCIENCE)杂志在线(online)发表题为 “A G protein-coupled receptor is a plasma membrane receptor for plant hormone abscisic acid” 的论文,该论文报道一种G蛋白偶联受体是植物激素脱落酸(Abscisic acid,ABA)的受体,该受体通过与异三聚体G蛋白α亚基直接相互作用传递脱落酸信号并调控脱落酸众多反应。

脱落酸是一种重要的植物激素,它参与调控众多植物生长和发育过程,特别是在植物对逆境,如干旱、盐碱、低温等的反应中发挥重要作用。ABA受体的研究对了解ABA的作用及其机制有非常重要的意义。以前的众多研究结果表明ABA的作用位点在质膜上,而且这种推测的质膜上的ABA受体可能是ABA参与逆境反应的主要参与者。

由G蛋白偶联受体(GPCR)介导的细胞信号转导途径是在真核生物中比较保守的信号转导机制。在动物细胞中GPCR介导的信号途径是重要的信号转导途径,比如动物对视觉、味觉、嗅觉的感知就是通过GPCR介导的信号途径实现的,动物中GPCR和G蛋白的种类也很多,它们参与的信号转导途径和生物学功能也很多。与此相反,高等植物中的GPCR和G蛋白的种类则非常有限,如模式生物拟南芥基因组中只有一种Gα,一种Gβ,2种Gγ和大约20-30种GPCR,而且到目前为止只有一种GPCR被比较详细地研究过,目前植物中还没有找到任何一种GPCR的配基。

该项工作鉴定了一种拟南芥中新的GPCR(GCR2),证实GCR2和Gα共同作用调控已知的ABA的众多反应。ABA可以与GCR2蛋白特异性的结合,而且这种结合符合配基-受体结合特性。研究人员进一步发现ABA与GCR2结合会导致GCR2-Gα蛋白复合体解离,从而释放出Gα使Gα进一步激活下游的效应器,从而证实GCR2是植物激素ABA的受体。该项工作的主要贡献有:1,鉴定确认了ABA的受体;2,明确了ABA的信号转导途径;3,发现了植物体系中的第一对配基-受体对。

该论文的第一作者是刘西岗博士,论文的其他作者还有岳艳玲、聂艳丽、李薇和中国农业大学的李斌和武维华博士。论文的通讯作者是马力耕博士。

2007 年2月16日,我所邵峰博士实验室在科学(Science) 杂志上发表题为 “The Phosphothreonine Lyase Activity of a Bacterial Type III Effector Family” 的文章。该文章报道了来源于多种三型分泌系统致病病原菌的一个效应分子家族,这类效应分子通过一种新的磷酸化苏氨酸裂解酶(Phosphothreonine Lyase)的活性,能够特异性地、不可逆地去磷酸化宿主MAPK激酶并使其失活。

通过所谓的三型分泌系统分泌效应分子进入真核宿主体内,进而阻断或调节宿主免疫相关的信号通路是许多革蓝氏阴性病原菌普遍采用的致病机制。寻找这些效应分子在宿主细胞中的靶蛋白并阐明其作用于靶蛋白及相关信号通路的生物化学机理对于我们认识病原菌致病机理和建立有效防治手段有着重要的意义。同时,这也可能促进我们对真核细胞本身的信号转导机制的进一步理解。 在这篇文章里,作者首先发现了一类高度保守的,既存在于动物病原菌[包括导致痢疾的志贺氏痢疾杆菌(shigella)和沙门氏菌(Salmonella) ],同时也存在于能够感染植物的假单胞杆菌(Pseudomonas syringae)中的三型分泌系统效应分子。通过对其中志贺氏痢疾杆菌来源的OspF效应分子的深入研究,作者发现OspF效应分子直接作用于真核宿主体内的,并且能够抑制MAPK激酶通路的激活。进一步的生化研究发现OspF具有全新的磷酸化苏氨酸裂解酶的活性, 这种酶活能够使OspF不可逆地“去磷酸化”MAPK激酶并使之失活。作者在文章中又证明了OspF效应分子家族都普遍具有这种MAPK激酶特异性的磷酸化苏氨酸裂解酶活性并探讨了可能的催化位点。

邵峰博士(本文通讯作者)说:“我们的发现不仅揭示了一种在动植物界都保守的致病病原菌拮抗真核宿主细胞免疫信号通路的机理, 但更为重要的是, 这种机理是由一种全新的酶学活性来完成的,并且突破了二十多年来关于蛋白质磷酸化和去磷酸化是可逆调节的认识”。

邵峰博士于1996年毕业于北京大学,2003年获美国密执安大学博士学位, 2005年7月应聘为研究所研究员并开始筹建实验室。此项工作从起始到论文发表,全部在研究所完成。对他在如此短的时间内所获得的进展,邵峰博士指出: “研究所为大家提供了一个最佳的科研环境。”

李洪涛博士,徐浩(技术员)和周艳(本科实习生)为本文共同第一作者;其他参与此项工作的还有,张杰(在读博士生),龙承祖(技术员),李素芹(技术员),陈涉(蛋白质组中心)和周俭民博士。
